研究專長:DNA 複製,修復,與重組,癌症與基因體不穩定機制。
研究方向:
癌症細胞具有基因體不穩定(Genomic instability)的特性,像是染色體數目非23對(aneuploidy),以及染色體重組(gross chromosomal rearrangement)的現象。其中基因體不穩定的現象常常是由於DNA複製(replication),修復(repair),與重組(recombination)機制的失調而造成的,因為這些機制的失調,而導致細胞會面臨不斷的複製壓力(replication stress),而造成染色體斷裂與重組。這些相關的調控基因的突變也常被發現在癌細胞中。像是同源重組(homologous recombination)基因BRCA1與BRCA2與范可尼(Fanconi Anemia)修復路徑基因的突變會導致基因體不穩定與癌症的生成息息相關。因此我的研究主要在探討DNA複製,修復,與重組相關的基因,其基因突變對複製壓力,造成基因體不穩定的分子機制。
我的實驗室已經開發許多研究複製壓力,重組,基因體不穩定的工具,包含DNA fiber, DNA combing, metaphase spreads, sister chromatid exchange, homologous recombination assay,iPOND以及SIRF。利用這些工具,我們可以研究基因突變對DNA複製的影響,包含replication speed,fork density,stalled forks, new origin firing, ssDNA replication gaps,染色體斷裂,重組,以及蛋白結合到複製叉的情形。這些平台也提供給國內外學者使用。
